Abstract:

Background & Objective: Helicobacter pylori has been recognized as a major risk factor in gastric and duodenum ulcers and gastric cancer. Some laboratory tests, such as culture, are not entirely satisfying. The aim of this study is to compare RUT with PCR in identifying H. pylori in the gastric biopsy tissue samples.Materials & Methods: First, the standard H.pylori sample (N:oC30) was provided. primers or the glmM (ureC) gene were used In PCR method The PCR test was optimized by sensitivity and specificity methods. Then 100 clinical samples composed of stomach tissue biopsy were prepared. The rapid urease test was conducted on all obtained samples to identify H.pylori. DNA was extracted from samples using DNG plus technique. Then, samples were studied using PCR method.  Results: The 294 bp product was amplified in the optimized PCR test. The PCR test sensitivity was obtained at 10 CFU. Any unwanted products were not seen in the specificity test with the exception of H. pylori DNA samples. Of 100 stomach biopsy samples, 64% were reported as positive by RUT and 76% by the PCR test. Conclusion: Given that the PCR test has higher sensitivity and specificity to detect H.pylori comparing rapid ureaas test, therefore this method could be used to detect H.pylori.

Compression of Histopathology, Culture and Rapid Urease Test in Diagnosis of Helicobacter Pylori in Gastric Biopsy Specimens

Abstract Background and Objective: Helicobacter pylori (H. Pylori) infection is related to chronic gastritis, peptic ulcer, duodenal ulcer and gastric cancer. Thus, identification and treatment of the infection have a considerable importance. The aim of this study was to compare three methods of Histopathology, Culture and Rapid Urease test (RUT) in identification of H. Pylori in gastric bio...

Determination of Helicobacter pylori Antigen in Stool Samples and Comparison with Rapid Urease Test in Patients Suspected of Helicobacter Infection

Background and Aims: Helicobacter pylori (H. Pylori) is the microorganism that infects nearly half of word's population. There are several invasive and non-invasive methods for diagnosis of infection. The main objective of this study was to evaluate antigen of H. Pylori in feces with H. pylori stool antigen (HpSA) test and comparison with rapid urease test (RUT) in the patients suspected to be ...

A low cost rapid urease test to detect Helicobacter pylori infection in resource limited settings.

The aim of the study was to evaluate the suitability of a modified one minute rapid urease test (one day rapid urease test) as a low cost H. pylori detection method. A sample of 205 patients clinically suspected of having H. pylori infection was tested. One day rapid urease test and histology based H. pylori tests (the gold standard) were performed on endoscopic antral biopsies. There were 6 tr...

Sensitivity and specificity of an in-house rapid urease test for detecting Helicobacter pylori infection on gastric biopsy.

We developed an in-house rapid urease test (iRUT) and evaluated the efficacy and the agreement of the iRUT and the cRUT compared with culture and histology for the detection of H. pylori infection. Five iRUT media were tested with H. pylori isolates and other bacteria. The most suitable iRUT medium was further evaluated for detection of H. pylori infection. Gastric biopsies from 120 patients we...
